It’s a pretty basic concept, but assigning a larger size to the buttons that you want to emphasize most is a great way to call attention to them (smart color choice is also key). Here are a few examples:
Are you referring to the dimensions of the buttons or their visual weight/emphasis? In the first example the button isn't that big really but you do see it right away.
I’m referring to both- a large button that blends into the background won’t be as effective as a large button that contrasts with the background, obviously. The dimensions, as well as the styling, are crucial elements.

I was thinking of the button dimensions. I was on a download site not two days ago and I had to scan the entire page twice to find the button to initiate the download – bah! – exasperating.

I think it depends on the context as well. Generally speaking, I like buttons to get my attention and prompt me to take an action. On a slightly separate note big buttons work well from Fitts law perspective as well because bigger the surface area, better the click through.
